سابقه و هدف: صرع یک بیماری مزمن است که تأثیرات متفاوتی بر عملکرد دارد. کنترل تشنج در کوتاه‌ترین زمان ممکن با داروهایی با کمترین عوارض جانبی مهم است. در ارتباط با اثربخشی و عوارض جانبی داروهای نسل جدید در مقایسه با داروهای قدیمی و خط اول بحث‌های زیادی وجود دارد. در این مطالعه اثربخشی و عوارض جانبی دو داروی سدیم والپروات (به‌عنوان داروی نسل اول) و لووتیراستام (به‌عنوان داروی نسل دوم) مقایسه شد.
مواد و روش‌‌ها: تمام کودکان بیشتر از 2 سال و کمتر از 18 سال با تشخیص جدید صرع عمومی و کانونی که معیارهای ورود و رضایت شخصی را برای ورود به مطالعه داشتند، به‌طور تصادفی وارد مطالعه شدند. بیماران به دو گروه تقسیم شدند؛ یک گروه با والپروات سدیم و گروه دیگر با لووتیراستام تحت درمان قرار گرفتند. فراوانی تشنج‌های مکرر در بیماران و عوارض بعد از مصرف داروها با پیگیری به مدت شش ماه به‌طور دقیق ثبت شد.
یافته‌ها: نتایج نشان داد بین سن، جنس و سابقه خانوادگی تشنج در بیماران هر دو گروه با میزان موفقیت در کنترل تشنج تفاوت معنی‌داری وجود ندارد. همچنین بین نوع داروی مصرفی (والپروات سدیم و لووتیراستام) و نوع تشنج (کانونی و عمومی) با موفقیت در درمان (مهار تشنج) بین دو گروه تفاوت معنی‌داری وجود نداشت. نتایج نشان داد گروه تحت درمان با والپروات سدیم عوارض جانبی بیشتری نسبت به گروه لووتراستام داشتند (0/05P<).
نتیجه‌گیری: با توجه به نتایج مطالعه حاضر، والپروات سدیم و لووتراستام در کنترل تشنج مؤثر هستند. برای برداشتن مراحل مؤثر برای کنترل هزینه‌ها و عوارض درمان ضدتشنج، پیگیری بیشتر و مطالعات بیشتر لازم است.
