Structural and Functional Investigation of activated form of blood coagulation factor X

<p><strong>مقدمه:</strong> فاکتور انعقادی X یک پروتئین مهم در مسیر انعقاد خون است. این فاکتور دارای ویژگی های ساختاری مهمی است که عملکرد این پروتئین را تحت تاثیر قرار می دهد. هدف از این مطالعه بررسی شاخص های ساختاری-عملکردی فاکتورX فعال در حضور یون های کلسیم بود. فاکتور X دارای 4 دومین است که دومین گاما کربوکسی گلوتامیک اسید (GLA) دارای آمینواسید هایی با بار منفی و همچنین آمینواسیدهای آب گریز است. این دو گروه آمینواسید با ویژگی متضاد، نقش اتصال به یون های کلسیم و غشا را به ترتیب برعهده دارند و پیچ خوردگی سه بعدی مناسبی را در حضور یون های کلسیم به فاکتور X می دهند. دومین های شبیه به فاکتور رشد اپیدرمی (Epidermal growth Factor) که شامل EGF1 وEGF2 می باشد، نیز رابط بین دومین GLA و سرین پروتئاز (SP) می باشند و در اتصال به کلسیم نیز نقش دارند.</p>
<p><strong>روش کار:</strong> روش گردآوری و جمعبندی اطلاعات از پژوهش مولکولی انجام شده در این زمینه بوده است. که شامل توصیف ویژگیهای ساختاری و سه بعدی فاکتور X بوده است.</p>
<p><strong>نتیجه گیری:</strong> نقش کاتالیتیکی فاکتور X برعهدۀ دومین سرین پروتئاز می باشد که این دومین زنجیرۀ سنگین فاکتور X را تشکیل می دهد و آمینواسید های این دومین در اتصال فاکتور X به مهار کننده و فعال کننده های مختلف نقش دارند.</p>
<p><strong>Introduction:</strong> Coagulation factor X is an important protein in the blood coagulation pathway. It contains significant structural features that affect its function. The purpose of this study was to investigate the structural-functional features of activated form of Factor X in the presence of calcium ions. Factor X consists of 4 domains. Gamma-carboxyl glutamic acid (GLA) domain contains negatively charged and hydrophobic amino acids. These two amino acid groups with contrasting characteristics are responsible for binding to calcium ions and the membrane respectively, so it provides appropriate folding of Factor X in the presence of calcium ions. Epidermal growth factor-like domain (Epidermal growth Factor) which includes EGF1 and EGF2, is the edge between GLA and serine protease domain (SP) and it is involved in binding calcium.</p>
<p><strong>Methods:</strong> Gathering and collecting of data has been done from molecular research in this area. It includes descriptions of three-dimensional structural characteristics of the Factor X.</p>
<p><strong>Conclusion:</strong> The catalytic role of Factor X is performed by SP domain which made the heavy chain of factor X. Amino acids of SP domain play role in binding factor X to different activator and inhibitor.</p>
